Abstract

Coconut tissue culture is an important technology for the future of the coconut industry. This paper has reviewed many valuable studies in the recent four decades. The importance of coconut tissue culture, differences and advantages of varies of explants (plumule, inflorescence, ovary), the influence of different medium compositions and environmental conditions were presented in detail. A brief summary of many protocols created worldwide and China has been made. Subsequently, the restrains of coconut tissue culture were identified and listed, and some suggestions were made to overcome the limits. This literature review could help the coconut tissue-culturists to further discover the potential of coconut tissue culture.

椰子(Cocos nucifera L.)属棕榈科椰子属,是一种高经济价值的多年生单子叶乔木[1]。作为一种热带油料作物和食品原料,椰子在93个国家和地区均有分布[2]。目前,椰子种植业面临很多严重的威胁,如生物性和非生物性胁迫及市场不稳定等[3]。作为最难以在体外繁殖的植物之一,传统营养繁殖手段,如扦插、压条、嫁接等很难用于椰子的培育繁殖。但是,利用组织培养技术,通过多种外植体诱导能够批量获得椰子的再生苗。本文总结了近40年(1980—2020年)国内外椰子组培工作的研究现状及存在问题。

1 椰子及其重要性

椰子是棕榈科椰子属中唯一的物种[4]。很多学者认为椰子源自于波利尼西亚或者印度群岛[5]。作为一种经济作物,椰子遍布世界热带地区,在全球超过90个国家均有分布[6]。生长在不同地域的椰子,具有多样遗传学特性[4]。椰子是满足热区人民食品和油料需求的重要生存资源[7],也被称为“生命之树”“大自然的超级市场”或“天堂之树”[8],在全世界热带地区尤其是亚太国家民生保障和食品安全中具有重要作用[9],每年有大量的椰果及相关加工产品进入世界商品市场[8]。据世界粮农组织(FAO)2016年统计,世界椰子总产量为5900万t,其中91%产自东南亚地区[10]。2014年,仅印度尼西亚就生产椰子1800万t[9]。在菲律宾,椰子产业的农业人口从业比例和农业用地为各行业内第一[11];在印度,有1200万人从事椰子种植、加工或销售产业[9];在马来西亚,超过90%的椰子种植户是风险抵御能力相对较弱的中小型农户[12]。由此可见,椰子生产对各椰子生产国的社会稳定、人民生活和经济发展都有着巨大的影响。

2 椰子组培——椰子产业潜在的“助推器”

目前,世界椰子产业面临着严峻的挑战。斯里兰卡出口发展委员会在2018年预测,2020年之前,世界椰子市场规模的增长幅度可达27%[13],相应椰果等加工原材料的需求也将大幅度增加。但椰子产量自2010年就处于平台期,未出现明显增长[14],这表明椰子供需平衡的局面被打破。而产量高、品种优良的椰苗将是椰子产业可持续发展、椰农收入提升的关键[15]。目前每年生产的椰苗只能达到市场需求的20%,种植户需要更多的种苗用于更新老化和遭受病害的椰树[16]。传统椰苗培育方法使用种果繁殖,繁殖系数较低,异花授粉导致后代性状分离而个体差异大,无法保持亲本优良性状;种果体积大,休眠期短,难以大批量远距离运输。而椰子茎干孤立,顶芽为营养芽,侧芽分化为花序,一般无分枝,因此扦插、嫁接、高空压条方法无法应用于椰子的无性繁殖[2]。椰子从种子到第1次结果往往需要5~6 a[17],最长可达8 a[18]。因此,传统椰子繁殖方式无法持续提供稳定高产优质的种苗,更无法为世界椰子产业的发展提供足够的动力[19]。为了弥补此劣势,组培快繁技术被应用于椰子繁殖并快速发展[17]。过去60年里,椰子组培研究集中于胚培养(embryo culture)、体细胞再生(somatic embryogenesis)、花药培养(anther culture)以及种质的低温保存(cryopreservation)和遗传转化(genetic transformation)[3]。成熟的椰子组培快繁技术可在较短时间内,以较低的成本培育大量性状优良或抗性强的椰子品种,从而提升椰农的收入[3, 19]
椰子组培的目标是获得能够发育成完整植株的体胚,而体胚发生途径是一种适用于与母体无维管连接的非合子细胞(组织)的组培方法,通过培养可形成具有两极性、类似合子胚的组织结构[4, 20]。体胚发生途径如下(见图1):外植体启动、愈伤组织诱导、胚性愈伤组织生成、体胚诱导及萌发[21]。与传统种苗生产、胚培养等方式不同的是,利用组培技术,一个外植体理论上最多可产生98 000株椰子种苗[4]。早在20世纪70年代,科学家就认为椰子体胚发生途径是一种可行的试管椰苗生产方式[18]

3 椰子组培简史

在科研人员的不断探索中,椰子组培技术在40年内快速发展。20世纪70年代早期英国Wye College第1次尝试椰子组培后,多种外植体被用于椰子组培中,具体分为体细胞组织和合子组织。其中,体细胞组织的外植体包括嫩叶、未成熟花序、根、顶端生长点等,合子组织包括未成熟/成熟合子胚和胚芽[22,23]。此外还有子房、花药等其他外植体[24]。20世纪八九十年代,上述外植体开始被用于椰子组培[25],其中以完整胚、胚芽、花序、子房为诱导材料的较为常见,但试验重复性不佳[16, 26]。1998年之前,大多使用花序作为诱导愈伤组织的外植体[27,28]。Blake等[29]和Hornung[30]最早使用椰子胚芽,但在得到胚性愈伤组织后未见后续报道[31]。1998年,墨西哥科学家报道了以胚芽为外植体获得体胚的具体操作方法,该项技术使得从整胚中提取的胚芽成为最适用于产生胚性愈伤组织和体胚的外植体[30]。科学家们意识到作为一种分生组织,胚芽比未成熟花序对于诱导条件更敏感、诱导效率更高[23, 30]。此后,研究者从操作方法、基础培养基成分、植物生长调节剂如生长素、油菜素内酯[32]、赤霉素[25]等方面对该方法进行了改良。尽管椰子组培研究起步较早,但由于椰子的体细胞组织对于试管培养环境的敏感性较差,取得成果有限,使得椰子成为最难以通过实验室方法生产的植物之一[16, 33-34],目前尚未发展出能够被普遍接受的组培技术[22]。因此,开发更高效率的椰子组培技术,尤其是探索通过体胚发生途径生产大量优良品种椰子苗的技术是非常必要的[4]。椰子组织培养时间线见表1图2

4 体细胞培养体系

4.1 以花序为外植体

目前认为,体细胞及其组织可产生保持亲本性状的后代。未成熟花序及其花序轴(rachilla tissues)是早期椰子组培的常用外植体[25]。早在1977年,Eeuwens等[35]就尝试使用椰子花序穗轴和幼嫩花序,并首次使用了Y3培养基。花序穗轴和幼嫩花絮经过诱导后产生的“胶质体”(colloids)去分化后形成愈伤组织[65],最终形成体胚并在适宜环境内萌发形成完整幼苗[27, 66]。早期,使用穗轴的诱导方法再生能力有限,效果相对较差,经过培养基组分及激素浓度的改良后诱导效率提高[27]。Verdeil等[27]使用含有0.2%活性炭和(1.5~3.5)×10-4 mol/L 2,4-D的固体培养基[18]。Magnaval等[67]以MS培养基中的大量元素、Nitsch培养基中的微量元素、MW培养基中的维生素组成的改良基础养基,添加2,4-D后诱导获得愈伤组织。Vidhana等[68]尝试了4种不同的培养基,在含有24 μmol/L 2,4-D和0.25%活性炭的培养基中获得了愈伤组织。
Perera等[34]认为,可用于诱导愈伤组织的花序的最佳取样时间难以判断,且取样过程可能会对树的茎尖造成致死性损伤。此外,花序的愈伤组织诱导率通常低于30%,实验可重复性差[30]。Vidhana等[68]的研究中约有30%的花序产生了高度胚性的愈伤组织,但诱导出的椰子组培苗茎尖无顶端分生组织,部分组培苗虽长出花枝,但有发育不正常的小花[1]。国内学者在试验过程中也发现了该现象[1]。因此椰子未成熟花序作为外植体存在诸多限制。

4.2 花药、子房作为外植体

20世纪80年代至今,Kovoor[69]、Iyer等[70]、Thanh-Tuyen等[56]、Monfort[71]等科学家尝试诱导花药,但均未获得完整植株。Thanh-Tuyen等[56]在Blaydes培养基中加入6%~9%蔗糖,15%椰子水,0.5%活性炭以及2 mg/L NAA,体胚形成率低于1%[72]。Monfort[71]利用花粉获得了少量的体胚,体胚萌发形成根尖和叶原基,但未发育成完整植株。Perera等[73]将产生的愈伤组织依次转接至含有66 μmol/L 2,4-D的Y3培养基,无激素Y3培养基和含有5 μmol/L BAP和0.35 μmol/L GA3的Y3培养基进行诱导,通过100颗花药产生的125个愈伤组织或胚性愈伤组织,获得了20株组培苗[49]。同时Perera等[73]也发现kinetin和2iP可以提高愈伤组织的发生率。
子房源于亲本体细胞,具有独特的优势。Perera等[50]以椰子幼嫩雌花中未受精子房为外植体,在含有100 μmol/L 2,4-D和0.1%活性炭的培养基上获得愈伤组织,诱导率为41%,愈伤组织在含有5 μmol/L ABA的培养基上连续继代培养后产生体胚。Perera等[50]还发现体胚在含有5 μmol/L BAP的培养基中可正常萌发形成芽。进一步试验表明,当子房外植体接种在CRI72培养基(含有100 μmol/L 2,4-D和0.1%活性炭)时,可以稳定地产生愈伤组织;9 μmol/L TDZ可提高愈伤组织诱导率;从愈伤组织转变至体胚的过程需要GA3激活;含有5 μmol/L 6BA和2iP的培养基能够增加体胚成苗率。产生的体胚可继代于含有66 μmol/L 2,4-D的培养基,但使用Y3培养基成长的体胚不需要任何植物生长调节剂也可以正常生长发育[50]。目前已建立较为完整的椰子子房再生体系,多种植物激素如TDZ、ABA、2iP、GA3被用于诱导试验中[50],效果不一。Perera等[50]利用该体系诱导出83株完整植株。Bandupriya等[75]根据前人经验,归纳出完整的椰子子房再生体系,试验流程见表2

经过数十载的努力,椰子花药、子房作为一种可行的组培外植体,拥有了较为完整的诱导体系。但相比之下,以花药和子房作为外植体,只能获得少量组培苗,繁殖系数较低[35],远不能满足商业需求[75,76]

4.3 其他体细胞作为外植体

已有研究表明,种苗或成熟椰子树上的幼叶均可诱导出胚性愈伤组织[36]。以5 a树龄的椰子树幼叶为外植体,最短可在6个月内生成胚性愈伤组织。但剥取幼叶会对树体产生破坏,同时愈伤组织形成率非常低(低于20%)[36, 77],现已不再使用。

5 胚和胚芽作为外植体

外植体中较为成功的有穗轴(未成熟花序)、花药、未受精子房,但在产生愈伤组织和体胚方面,胚和胚芽优于其他几种外植体[23, 30, 74]。根据Pérez-Núñez等[4]的计算,使用一个胚芽理论上可产生98 000个体胚,与直接使用体细胞进行培育相比,其效率高出约五万倍。因此,椰子胚芽使得开发更高效的组培技术成为可能[23, 78]。使用胚芽进行椰子无性繁殖多选用具有优良表现型(抗病等)的亲本[23]。近10年,学者将椰子无性繁殖的研究重点逐渐由难以操作的体细胞培养转向简单易行的合子细胞培养[3]。目前,愈伤组织诱导、体胚形成、体胚萌发和成苗等已成为研究热点[25]
合子胚的成熟度是影响愈伤组织诱导、体胚生成的重要因素。5~6月龄的胚愈伤组织诱导率低,但8月龄及以上的胚多已萌发,无法使用[40]。未成熟合子胚的胚性愈伤组织诱导率约为50%,远高于成熟胚的3%[25]
植物激素和植物生长调节剂等会显著影响椰子组培的效率。Karunaratne等[40]使用开花后6~7个月的胚,在12~20 μmol/L 2,4-D诱导下可产生白色、完整的、可产生体胚的愈伤组织,继续使用8 μmol/L 2,4-D诱导,一半的愈伤组织产生了球状体胚,继而在10 μmol/L kinetin和BAP的刺激下,22%的体胚萌芽。除此之外,渗透压调节物(osmoticum)、ABA、GA3对于椰子胚的萌发和成熟也有一定作用[79,80]。使用与Karunaratne等[40]相同的方法诱导愈伤组织后,Fernando等[46]使用2.5~7.5 μmol/L ABA诱导,在3~7周后可得到体胚。该实验认为含有ABA的培养基可诱导大量胚性愈伤组织、嫩芽以至完整植株,其质量甚至高于在低浓度2,4-D培养基中得到的植株。
在使用椰子胚芽作为外植体进行无性繁殖时,2,4-D可以快速诱导出愈伤组织。Hornung[30]使用400 μmol/L 2,4-D,约一年后得到成熟体胚。油菜素甾醇及抗细胞分裂素(anticytokinins)也可以使体胚诱导率增加1.5~2.5倍[32]。Rajesh等[16]在诱导初级愈伤组织时使用4.54 μmol/L TDZ,在继代培养时使用100 μmol/L精氨(spermine)和1000 μmol/L腐胺(putrescine),结果显示外源多胺类可以提高椰子体胚发生率。
早期,科学家多使用整胚(成熟或不成熟的)作为椰子组培的外植体。此后,尝试胚不同部位的切片用作外植体[45, 81]。后期验证,从胚的中心取得的切片最适于产生胚性愈伤组织,诱导率为58%,当使用远离胚中轴的组织时,愈伤组织的诱导率会急剧下降[25]。随着时间的推移,胚切片技术已不再用于愈伤组织的诱导,取而代之的是直接从胚中分离胚芽[47]。目前掌握该技术的研究机构主要有澳大利亚昆士兰大学(The University of Queensland)和墨西哥尤卡坦科学研究中心(Unidad de Biotecnología, Centro de Investigación Científica de Yucatán, CICY)等。该技术可对次级体胚(secondary somatic embryos)进行数次重复分割并继代以增加体胚的产量,尤其对组培低敏感型的植物(例如椰子)来说是快速增殖的方法之一[82]。几十年内,以胚芽为外植体的椰子组培快繁技术快速发展,并被广泛使用。但使用椰子胚芽作为外植体也有限制,例如异花传粉导致胚芽可能为杂合子,从而诱导产生的组培苗表现型无法确定,可能出现与亲本性状不一致现象而产生后代异质性(heterogeneity)[74]。此外,该体系还存在组织褐化、反应不敏感、体胚诱导率低、个体特异导致的组织变异等问题[16, 26]

6 中国椰子组培研究进展

相比世界椰子主产国,中国椰子组培快繁技术起步较晚。究其原因,椰子并非中国的主要经济作物,全国只有海南省能够规模化、商业化生产椰果,因此国内市场动力不足,研究投入较少,椰子产业的发展相对滞后,所以与世界椰子主产国相比,椰子相关的科学研究也较少。
国内椰子组培研究最早报道于1991年。陈幸华[83]以3%蔗糖和0.5%琼脂的MS培养基为基础培养基,加入NAA、KT、BA和2,4-D和不同剂量的活性炭研究椰子胚的离体培养,试验结果表明,活性炭不仅能防止褐变,还可以提高成苗率。1997年,国内学者开始探索椰子幼嫩花序离体培养的器官发生途径,诱导幼嫩花序产生愈伤组织,后期花序轴的根部长出嫩芽,继续培养后形成了具有根、叶结构的完整植株[1]。在以离体胚为外植体进行培养的过程中,研究发现5%蔗糖浓度的培养基最适于进行椰子合子胚的离体培养[84]。2010年,中国热带农业科学院椰子研究所以椰子成熟胚和幼嫩花序为诱导材料研究了愈伤组织诱导及其褐变的控制,研究结果表明长度为16.5 cm佛焰苞(幼花序)诱导效果最好;含有25 mg/L 2,4-D、2.5 g/L活性炭和5 g/L聚乙烯吡咯烷酮(PVP)的Y3培养基的诱导率最高且对材料褐变有抑制作用[85]。在愈伤组织诱导和体胚诱导中,通常以添加2,4-D、麦草畏、NAA等激素的Y3培养基为诱导培养基。2014年,国内学者使用布迪椰子(Butia capitata或冻子椰子)种子成熟胚和含有0.5 mg/L NAA,0.5 mg/L 2,4-D,0.5 mg/L BAP,60 g/L蔗糖,6 g/L琼脂的MS培养基,诱导出的愈伤组织效果最好[86]
综上所述,国内的椰子组培快繁研究起步较晚。从研究的主体来看,主要集中于培养基的研究,缺乏对于椰子组培快繁的全过程研究以及基于组培技术的转基因技术研究;从外植体的角度来看,主要采用比较简便易行的体细胞培养(如花序)或整胚作为外植体进行试验,缺乏使用主流高产外植体如胚芽进行研究。国内的椰子无性繁殖研究与此领域内的领先国家,如墨西哥、澳大利亚、印度尼西亚、越南、印度、斯里兰卡等国仍有明显差距。

7 存在问题与展望

7.1 存在问题

40年来国内外对椰子组织培养进行了大量的研究,在理论、操作等方面取得了进步并初步建立了组培快繁体系,但尚未成熟,未被广泛推广和应用[2]。目前还有很多部分解决或未解决的问题,例如外植体对于组培的响应不同、试管苗生长缓慢、移栽田间的试管苗缺乏活力等[3]。Gichner等[87]认为组培技术无法推广的原因主要有以下五点:技术不成熟、时间长且成本高;长期无法解决工程学、生物学对组培的限制;无性繁殖植株质量极低;复合人才缺乏。针对椰子组培来说,有以下技术难点:首先合子胚的培育效率较高但合子胚多为杂合子,无法在早期确定组培苗的性状,需等待数年至结果才可观测;其次亲本基因型、人员技术、内源及外源污染、生长微环境变化、组织褐化、不成熟的炼苗技术均对椰子组培的结果产生巨大影响,损失率高;技术重复性较差,实验结果差异大。不同批次的椰果、试剂、操作人员、操作环境等因素的差异都有可能导致试验无法重复。同时其他棕榈类组培结果显示,组培苗移栽大田后可能会出现大量未知变异。凭借目前的技术,此类变数在组培阶段无法预测。
在椰子组培技术的回报率与商业化推广目前尚不明确的情况下,如何推广现有成熟的椰子组培技术(如胚培养技术)是目前行业存在的主要问题之一。当下组培技术成本较高,对于发展中国家更是如此。椰子十大主产国为印度尼西亚、菲律宾、印度、巴西、斯里兰卡、越南、巴布亚新几内亚、墨西哥、泰国和马来西亚,这些国家多为发展中国家,科研水平较低、科研投资较少,部分国家不具备独立完成椰子组培工作的软、硬件实力和经济基础。商业组培的成本主要包含组培实验室建筑及内部布置费用、试剂耗材费用和基本的水电运行费用。国际原子能机构(IAEA)与世界粮农组织(FAO)对印度调研后发现,在印度设立一家低成本、中等规模(可生产储存20万株左右的组培苗)的组培工厂(195 m2),土地费用和改建费用可达6.25万美元,设备租用费为5000美元/年[88]。因此组培技术只能用于具有高价值的植物种类[89]。一些发展中国家的学者采取试剂替代品希望能在硬件方面降低成本,如Bhattacharya等[90]采用2种印度常见植物西谷椰子(Metroxylon sagu Rottb.)和卵叶车前(Plantago ovata Forsk.)的提取物作为琼脂的廉价替代物,同时使用滤纸、尼龙布、聚苯乙烯泡沫(polystyrene)和玻璃棉(glass wool cloth)作为支撑材料。Shivakumar等[91]提出使用壳聚糖(chitosan)作为一种低成本抗菌剂以减少内生菌的污染。但对于成本占比最高的人工成本(如技术员、劳工、检测员、销售市场人员)及人员招募、培训费用目前仍无有效的解决办法[92]。Gichner等[87]也认为在组培过程中转移体胚、嫩芽、植株等植物组织的步骤对实操技术要求较高。不仅如此,目前组培技术商业化产生的高额运营成本[89]也导致该技术无法广泛使用。

7.2 未来展望

包括生物反应器(bioreactor)在内的各种新型生物技术已经广泛运用于植物的组培快繁技术,但是目前在椰子组培中还未见报道。Nguyen等[3]认为,未来的椰子组培研究方向应着重于改良培养基和建立悬浮培养体系(suspension culture system),以提升生产体胚的效率,同时在炼苗步骤中也应更多地关注间歇浸没系统(temporary immersion system)与光能自养系统(photoautotrophic systems)。目前已出现一些专门针对植物组织培养的生物反应器,如筏式/喷雾式生物反应器,其中间歇浸没系统使用较为广泛[93]。与固体、半固体、液体培养相比,间歇式浸没培养可提高繁殖系数,同时减少劳动力、提高试管苗质量、增加驯化成活率,但其劳动力成本仅为固体培养的一半[94],总成本可降低20%[95]。将组培苗从试管环境中转移至室外环境是劳动密集型的工作,使用大批量生产技术可减少12%的成本支出,使用改良的组培方法可以减少24%,而使用半自动设备的成本降低幅度可达35%[87]。Nguyen[3]认为,在未来可以尝试使用含有多种有益成分的“液体胚乳”——椰子水作为液体培养基。同时,可通过研究体胚生成的相关基因来实现椰子组培技术效率的提升。通过了解椰子体胚发生基因的作用机理,可以筛选出更适于椰子组培的亲本材料以减少组培工作对于椰子种果的需求。
国际合作在椰子组培以及相关领域的作用越来越重要。通过各国间的椰子种质资源互换和组培技术交流,椰子组培技术在全世界很多国家生根发芽。目前墨西哥尤卡坦科学研究中心(CICY)、澳大利亚昆士兰大学(The University of Queensland)掌握了使用胚芽诱导愈伤组织生成植株的核心技术;法国农业国际合作研究发展中心(Centre de coopération internationale en recherche agronomique pour le développement, CIRAD)在胚培养技术以及生物反应器等方面处于领先;斯里兰卡椰子研究所(Coconut Research Institute of Sri Lanka)则掌握了以子房和花药为外植体的椰子组织培养的核心技术。通过与以上这些研究机构的学术交流,可以增强椰子组培技术的通用性。目前包括中国在内的39个椰子生产国加入了国际椰子遗传资源网(International Coconut Genetic Resources Network, COGENT)致力于椰子产业的可持续发展以及椰子繁殖技术的推广。包括椰子组培在内的国际技术合作已经引起了各国的重视,将来国际合作一定会为椰子组培事业带来全新的动力。
